Quick question on the hydrogel solution- PFA fixation concentration

We are using CLARITY to visualize lungs/lymph nodes tissue. The lab uses a lower concentration of PFA when fixing the lymph node/lung tissue. I'm wondering if its necessary to use the recommended 16% PFA. I feel that the protocol works best for brain tissues rather than other organs.

  • What is the concentration of PFA that you normally use? Keep in mind that the final concentration of PFA in the hydrogel solution is actually 4%. The PFA is essential in attaching the acrylamide monomers to all the biomolecules (proteins, DNA, etc) in the tissue, so you may not want to lower the concentration.
