        <description><![CDATA[Holding the tubes onto the reservoir tank can be a pain. Tape stinks; it leaves marks, doesn't hold well, and it is difficult to control the depth of the inlet and the outlet (typically you'll want the inlet to be below the outlet to minimize bubbles). So I remixed https://www.thingiverse.com/thing:1212828 to add holes for 0.5" tubing. Clip it onto a beaker and insert the tube through one of the holes (choose an angle/side). FDM print with whatever material (nGen preferred), no supports, 20% infill, and high resolution (Cura) if you can afford the time. More projects can be found: https://optogeneticsandneuralengineeringcore.github.io/ONECoreSite/ (including CLARITY) and I may put this up there some day (doubtful). Don't have a printer? I am a first year master student, and I'm attempting to use CLARITY as a tool in my master thesis. I am currently scavenging for supplies. Before I order FocusClear, however, does anyone know how much is usually used per experiment? I looked through the protocol, but I can't see that it says how much  you're supposed to use. Just a rough estimate so I know how much I should order.

RapiClear 1.47.

The datasheet says its a water-soluble clearing reagent. It also suggest it can be applied for 
techniques like CLARITY. They even show some images of different mouse tissues in RapiClear, with pretty
decent clearing.

Does somebody already tried this RapiClear on his/her samples

        <description><![CDATA[I just did a CLARITY setup and have done my first clearing of kidney tissue. It all works fine, except for problems with the electrodes. I use 0.25mm palladium electrodes and these burn out after just a couple of days of clearing, even if running at a fairly low voltage (10V). Did anyone have the same problem with platinum electrodes? Do you think the problem is the thinner wire or that I´m using palladium as electrode material? To my understanding platinum and palladium should have similar properties in terms of reactivity and resistivity.]]>

I'm looking to test the clarity method on some knock in fluoro reporter mice and some human neural tissue.

The CLARITY method may not be suitable for my tissue, and so I am trying to not set up the entire system here to then discover it's useless.

Could anyone possibly spare a minimal amount of time to allow me to visit and try their system on my tissue? I would obviously reimburse you for any reagents etc.

I'm based in Edinburgh, and can travel anywhere in the UK or even to Europe!

        <description><![CDATA[I noticed in the brains I've cleared there has been a white precipitate forming when placed in Focus Clear, and when viewed under the microscope there are very noticeable holes in the tissue. We think it may be the result of the way we made the clearing solution. When making the solution, we just added the boric acid and sds, followed by the dH2O. A white precipitate formed, and it took more than 200 pellets of NaOH (for 10L of clearing solution) to bring the pH to 8.5.

Has anyone had this issue, or is there a specific protocol that works better for making the clearing solution (like dissolving either boric acid or sds first?). Thanks! ]]>

We are setting up our CLARITY system now, but we are having problems sourcing a filter similar to that used in the original paper (McMaster Carr doesn't ship outside of the US it seems).

Has anyone used a different filter to what was specified? Do we need the filter, other than clear debris that might otherwise get stuck in the ETC? Any suggestions or thoughts on this would be welcomed!

There doesn't seem to be any easy way to find out how to order it or how much it actually costs.
