well using the lamp you will only see autofluorescence. Sometimes if your signal is really strong you are able to see it. For me the best option is to use the binocular to search for an area in the tissue where I know there should be signal, then ch…
normally I do the cutting after hydrogel polymerisation.
I work with brain so its the best moment to use a brain matrix.
If you don't need to be very specific about the region you will cut out, you can also do it after the clearing with a sharp…
Dont`t put samples that are in clearing solution in temperatures below RT. If you do this the SDS will precipitate. If you want to store the samples PBST is good, for long-term 4° PBST + diluted azide works fine for me
I dont know from which country you are but here is a supplier for Europe.
You should get all parts you need
but los for some lab supplier, electronic store or DIY store they also have this kind of stuff
its normal for samples to swell in Clearing solution. I never used glycerol only FocusClear.
For me the samples return to their original size.
As far as I know FocusClear gives better results in comparison to glycerol. The hydrogel likes mor…
I can quickly answer maybe your 1st question.
I had also a batch of SDS that gave me a white clearing solution.
I called the company and got a new one (Sigma)
With the new batch the clearing solution was clear as always.
So I would not risk using…
If you want to check how clear it is:
Wash with PBST then incubate in FocusClear.
If the brain is clear then wash again with PBST and continue with antibody staining.
If its not clear then wash again with PBS, then change to clearing buffer and co…
Hi I have not noticed white precipitate nn my sample. It could be SDS accumulation. It happens when the clearing buffers dries out, you get white SDS marks. Try changing the flow rate and checking that your sample is always fully in the solution. Al…
if I recall correctly they FedExed it to me, went trough customs before but no big problem there, but this was like 5 months ago. Maybe they changed their policies now.
I saw that RapiClear is a newer product, came after CLARITY. So maybe is more o…
I think these all depends on your settings. Which resolution, how big is the scan area, which kind of objective, the frame average, what kind of stage do you have.
For me with my actual settings a 1.55 mm x 1.55 mm x 1.55 mm area takes 3h.
